Type 2 iodothyronine deiodinases (D2), which catalyze the deiodination of thyroxine (T(4)) into the more active triiodothyronine (T(3)), are proposed to play a crucial role in the regulation of thyroid hormone action in many peripheral tissues and in the brain. In the present study, we evaluated the regulation of D2 in the whole brain, hypothalamus, hippocampus, cerebellum, cortex, striatum, and in the brown adipose tissue (BAT) of rats following thyroxine (T(4)) and 3,5,3'-triiodothyronine (T(3)) administration. Following treatment with different doses of T(4) (0.02-2.0 microg) or T(3) (1-20 microg), D2 activity increased significantly in the brain. In addition, D2 activity was stimulated by both T(4) and T(3) in the BAT, in a tissue-specific manner. Thus, D2 activity increased following T(4) treatment, whereas T(3) treatment increased D2 activity in the BAT but had no effect on D2 in the brain. In contrast to D2 in the brain, D2 in the BAT was regulated by both T(4) and T(3), with a dose-dependent increase in D2 activity observed following both treatments. Additionally, the in vitro activation of brain D2 by either T(4) or T(3) was investigated, and the results of this study suggest that D2 in the brain and BAT is preferentially activated by T(3) in vivo. In contrast, in vitro activation of D2 by both T(4) and T(3) in the brain and BAT suggests the possibility that these D2 isoforms may be activated in a tissue-specific manner in vivo.
